Dimebon (latrepirdine) is a late-stage investigational clinical candidate for the treatment of two devastating neurodegenerative diseases, Alzheimer’s and Huntington diseases.
Overview
Dimebon is an investigational compound currently in Phase 3 development for the treatment of Alzheimer’s disease and Huntington disease—progressive, devastating neurodegenerative diseases. Dimebon has a unique mechanism of action, distinct from currently available treatments. In preclinical studies, dimebon has been shown to protect brain cells from damage and enhance brain cell survival, potentially by stabilizing and improving mitochondrial function.
We are currently evaluating dimebon with our partner Pfizer Inc in clinical trials for both Alzheimer’s and Huntington diseases. We have competed four trials with dimebon in Alzheimer’s and Huntington diseases and are currently enrolling patients in two additional placebo-controlled randomized trials.
Completed Trials:
- The first trial in patients with mild-to-moderate Alzheimer’s disease. Data from this trial have been reported and showed that dimebon had a statistically significant improvement over placebo on all primary and secondary endpoints.
- The Phase 3 CONNECTION trial in mild-to-moderate Alzheimer’s disease patients with mild-to-moderate Alzheimer’s disease. Data from this trial have been reported and showed that dimebon did not meet any of its co-primary or secondary efficacy endpoints, but was well tolerated.
- A placebo-controlled Phase 3 safety trial in mild-to-moderate Alzheimer’s disease patients on a variety of background anti-dementia medications. Data from this trial have been reported and demonstrated dimebon’s tolerability when dosed alone or in combination with approved Alzheimer’s disease medicines.
- A Phase 2 trial in Huntington disease. Data from this trial have been reported and showed that dimebon was well tolerated and significantly improved cognitive function in Huntington disease patients compared to those treated with a placebo.
Ongoing Trials:
- The Phase 3 CONCERT study, which is currently enrolling patients with mild-to-moderate Alzheimer’s disease who are already being treated with donepezil, the leading Alzheimer’s disease medication worldwide.
- The Phase 3 HORIZON trial, which is currently enrolling patients with Huntington disease to evaluate the potential benefits of dimebon on cognition in these patients.
